Retinoids have far more trial evidence than peptides for photoaged skin, and tretinoin is still the standard. Biomimetic peptides are small signaling or scaffold molecules with small, mostly maker-run trials, and most struggle to cross the skin barrier. Many people use both. We found no trial that compares retinol plus a peptide with retinol alone, or that tests which goes on first.
This page compares what each has shown, explains why delivery decides so much, and covers using them together. For the full guide to peptide types, see our biomimetic peptides page.
How to read it. Strong: randomized trial in people, on the stated outcome. Limited: small or maker-run trials. Very limited: one group or lab samples only.

What does each one do?
Tretinoin is the gold standard for photoaging. In a double-blind, vehicle-controlled trial, 14 of 15 treated faces improved and none of the vehicle faces did, and irritation was the main side effect (1). A 2024 review of 25 studies found comparators did better than tretinoin in 7, matched it in 13 and did worse in 3, and that most comparators were better tolerated. It also found that most of those studies were of poor quality or biased (2).
Retinol can come close. In a 3-month trial of 120 women, a cream with 0.2% retinol and a second ingredient (LR2412 at 2%) did not differ from 0.025% tretinoin on wrinkles, pigmentation or pores, and it was better tolerated (3).
Peptides work differently. Reviews group cosmetic peptides into signal, carrier, neurotransmitter-inhibitor and enzyme-inhibitor peptides (13).
Do peptides get into skin?
Mostly they struggle. A 2022 review concluded that most anti-wrinkle peptides are poor candidates for skin permeation, and that chemical modification, iontophoresis, microneedles, metal complexation or encapsulation can raise it (14). A 2025 review names poor permeability as the main drawback of cosmetic peptides (13). For acetyl hexapeptide-8, whether a cream can reach the nerve-muscle junction remains uncertain (6).
Newer lab work asks whether peptides must penetrate at all. In full-thickness human skin, palmitoyl hexapeptide-12 permeated the stratum corneum and epidermis, formed a gel using the skin’s own ions, and strengthened the barrier after the skin lipids were removed (12). A GHK peptide grafted with a cell-binding motif (RGD-GHK) reached a high local concentration in skin in permeation and adsorption tests (15). A designed collagen-mimetic peptide, TSTP-2, improved delivery by disturbing skin lipids (16). These are cell, skin-sample and animal experiments. None tested a finished product in people.
What is the best human evidence for peptides?
Palmitoyl pentapeptide-4 (Pal-KTTKS, the Matrixyl peptide) has the best-known trial. In 93 women aged 35 to 55, a moisturizer with 3 ppm of it went on one side of the face and the plain moisturizer on the other for 12 weeks. Wrinkles and fine lines improved more on the peptide side. The first author worked at Procter & Gamble (4).
Argireline (acetyl hexapeptide-8) at 10% in an emulsion cut wrinkle depth by up to 30% after 30 days in women volunteers (5).
P26, a peptide derived from a cornified-envelope protein, was tested in two double-blind, vehicle-controlled trials. The paper reports better barrier protection, firmness and roughness. The abstract gives no sample sizes (7).
A four-week study of 47 women used a lotion with 0.1% Tripeptide-29 plus niacinamide and two plant extracts. Hydration rose by up to 72.5%. The abstract describes a single group, and with four active ingredients the peptide cannot take the credit (11).
GHK-Cu has little trial data. In aesthetic medicine, 2 of its 20 studies are randomized trials, and a 13-person study after laser resurfacing was negative on objective measures (9, 10). Our GHK-Cu copper cream guide compares the creams.
Can a peptide replace retinol?
One trial says a peptide can beat it. Healthy volunteers used a 0.002% cyclized hexapeptide-9 serum, a 0.002% retinol serum or a vehicle serum twice daily for 56 days. The peptide reduced the number, area and roughness of crow’s feet and forehead wrinkles. Retinol reduced only some of them. The peptide did better on every outcome except crow’s feet roughness (8).
Read it with care. The retinol dose was 0.002%, while other trials here used 0.1% to 0.2% (3, 17). The peptide is a new design, and the abstract gives no sample size. This is one trial, and tretinoin still has the larger evidence base (2).
Can you use peptides and retinol together?
People do. In one 8-week study, 32 women with mild photoaging used an essence twice daily with 0.1% supramolecular retinol and 0.02% acetyl hexapeptide-1. Wrinkles, texture, elasticity, firmness and hydration improved, and it was well tolerated (17). All the women got the same essence, so the study cannot separate the two ingredients.
We found no trial that compares retinol plus a peptide with retinol alone, tests whether the order matters, or shows that a peptide reduces retinoid irritation. Advice on all three is opinion. Tolerability is the practical limit. The 2024 review says tretinoin’s poor tolerability often limits its use (2).
Sunscreen has stronger prevention data than either. In a randomized trial of 903 adults over 4.5 years, daily broad-spectrum sunscreen cut skin aging by 24% against discretionary use (18). Our best peptide for anti-aging page ranks the options.
How do you read a peptide product?
Each trial used a stated concentration. Pal-KTTKS was 3 ppm, acetyl hexapeptide-1 0.02%, Tripeptide-29 0.1% and cyclized hexapeptide-9 0.002% (4, 8, 11, 17). An ingredient list shows order, not amount, so a named peptide on a label does not match a trial. Delivery limits every peptide, and we found no study showing that a retail serum puts a useful amount into skin.
Peptides and retinol questions
Can peptides replace retinol?
Not on current evidence. Tretinoin has the larger evidence base and is still the standard. One 56-day trial found a novel cyclized hexapeptide beat a very low-dose retinol serum, and that is a single trial.
Which is better for wrinkles, peptides or retinol?
Retinoids have far more trial evidence. Peptides have small trials, mostly run by the makers, and most struggle to cross the skin barrier.
Can you use peptides and retinol together?
People do. One 8-week study of an essence with both reported better wrinkles, texture and firmness in 32 women, but it had no comparison group. We found no trial that compares the pair with retinol alone.
Should peptides go before or after retinol?
We found no trial that tests the order. Advice on it is opinion.
Do peptides penetrate skin?
Mostly they struggle. A 2022 review says most anti-wrinkle peptides are poor candidates without enhancement, and enhancement methods raise permeability in experiments.
Which peptides have the best human evidence?
Palmitoyl pentapeptide-4 has a 93-woman, 12-week split-face trial. P26 has two double-blind, vehicle-controlled trials, and GHK-Cu has two small randomized trials. Most trials are small and run by the makers.
Does retinol irritate skin?
Irritation was the main side effect of tretinoin in the key trial, and a 2024 review says poor tolerability often limits its use. A retinol cream was better tolerated than tretinoin in a 120-woman trial.
